Analysis of High-molecular-weight Glutenin Subunit Compositions from Hubei Commercial Hexaploid Wheat (Triticum aestivum L.) Cultivars

Abstract
Sodium dodecyl sulfate polyacrylamide gel electrophoresis was used to determine the high-molecular-weight glutenin subunit (HMW-GS) composition of 45 hexaploid wheat (Triticum aestivum L.) cultivars from Hubei Province.Of which 40 cultivars are homogeneous for HMW-GS composition and 5 cultivars are heterogenous.Nine alleles have been identified at Glu-1 loci and which are three at Glu-A1 (1, 2*, Null),four at Glu-B1 (7, 7+8, 7+9, 14+15) and two at Glu-D1 (2+12 and 5+10),respectively.The major HMW-GS at Glu-1 loci are Null,7+8, 2+12,with the frequencies of 62.5%, 60.0% and 72.5%,respectively.Twelve HMW-GS combinations have been observed,of which five combinations,(Null, 7+8, 2+12),(1, 7+8, 2+12),(1, 14+15, 2+12),(Null, 7+9, 2+12),(Null,7+8, 5+10) have accounted up to 72.5%,with each combination high than 10% in all and the first combination up to 30%.The Glu-1 quality scores of the tested cultivars range from 5 to 10 with an average score of 7.0 . The low frequency of subunits 5+10 implies that the analyzed cultivars having poor bread-making quality.
