Abstract
Oryza officinalis found in Guangxi Province, China is an important germplasm that possesses many valuable characteristics such as resistance to biotic and abiotic stresses.With biotin labelled total genomic DNA from O.officinalis as probe, unlabelled O.sativa total genomic DNAas blocking and HRP DABfor detection, we applied genomic in situ hybridization to somatic chromosome preparations of O.sativa × O. officinalis F1 hybrid.When blocking ratio being 1:20~30, 12 O.officinalis chromosomes were clearly identified from 12 O.sativa chromosomes.O.officinalis chromosomes appeared deep brown, but O.sativa chromosomes colored lightly only.In interphase nuclei, many spot like hybridization signals were detected, distributing mainly along nucleus side.
