The Application of the Image Deconvolution Technique to the Study of the Maize 45S rDNA Transcription Pattern

  • Due to the effect of the out-of-focus fluorescence,images gained by a wide-field fluorescence microscope were usually too hazy to be used in observing the fine structure of the plant nuclei.In this study,we discussed several important parameters which heavily affect the results of image deconvolution.After proper image deconvolution restoration,the images of the maize 45S rRNA in situ hybridization signals were clearly displayed.The analysis of the hybridization images showed that the transcription of the maize 45S rDNA gene was initiated in the dense fibrillar components(DFC),then transferred to the nucleolus inner space as the nucleolus transcription activity increased,and finally combined with the 5S rRNA to form the mature rRNA in the nucleolus cavity.The results showed that the application of the image deconvolution restoration technique could effectively improve the resolution of the two-dimensional image gained by a wide-field fluorescence microscope,and thus made the wide-field fluorescence microscopes more useful in observing the fine structure of the plant nuclei.
