Mitochondrial RNA Editing of ATPase atp6 Gene Transcripts of Yunnan Purple Rice(Oryza sativa L.)

Abstract
Base on the of construction of a new type of cytoplasmic male sterile line Zidao A(Oryza sativa L.),we analyzed the transcript editing of mitochondrial ATP synthase subunit 6 gene(atp6) from CMS line and its maintainer line.With PCR,RT-PCR,complete nucleotide sequences were determined for the mitochondrial atp6 gene cDNA from two lines of Purple Rice Type rice:cytoplasmic male sterile line Ying xiang A and its maintainer line Yingxiang B.The atp6 transcript of Yingxiang A had no editing site and the transcript of Ying xiang B had 16 editing sites with changes affecting 15 amino acid residues of the protein product.The editing of the atp6 transcript from Yingxiang B changed a glutamine codon(CAA) into a termination codon(TAA),shortening the protein of Ying xiang B to the “standard” size.The Ying xiang A transcript,which had no editing site and a“normal” termination codon,cannot be translated to a normal protein.The results demonstrated the important role of RNA editing in the production of the functional ATP6 subunit and suggested that RNA editing may be associated with cytoplasmic male sterility.
