Isolation of High Molecular Weight Glutenin Subunit Gene 1Dy12* from Yunnan Wheat(Triticum aestivum subsp.yunnanense King)

  • A novel y-type high molecular weight glutenin subunit(HMW-GS) possessing a slightly faster mobility than that of subunit 1Dy12 was identified through SDS-PAGE analysis,designated 1Dy12* in Yunnan wheat(Triticum aestivum subsp.yunnanense King).The coding region of HMW-GS gene 1Dy12* was isolated and sequenced with Glu-Dy locus specific primers.The complete open reading frame(ORF) of 1Dy12* was 1980 bp,encoding 658 amino acids.Comparison of amino acid sequences showed there were 3 amino acid differences between subunits 1Dy12* and 1Dy12 and a deletion of dipeptide(GQ) in subunit 1Dy12*.Also,there were fifteen amino acid differences between subunits 1Dy12* and 1Dy10,two hexapeptides(IGQGQQ) insertions and a deletion of dipeptide(GQ) in subunit 1Dy12*.This showed that subunit 1Dy12* is a novel HMW-GS,and it is in process of evaluating subunit 1Dy12* to effect wheat processing quality.
