Callus Induction from Immature Inflorescence and Plant Regeneration of Buffalograss

  • Female and male immature inflorescences of buffalograss Buchloe dactyloides (Nutt.)Engelm. were used as explants to develop a tissue culture system for callus induction,subcultureand plant regeneration. The result showed that it is more difficult to dedifferentiate to form callusfrom female inflorescences than male ones. Callus was initiated in induction medium containing2 mg/L 2,4-D from less than 8 mm male inflorescence,the frequency was 80.0%-86.8 %. Theinclusion of 10 mg/L AgN03 in medium had no stimulatory effect on the frequency of callusinduction,however it could improve callus quality. The combination of 2 mg/L 2,4-D with0.1 mg/L 6-BA contributed to callus subculture. The regenerating potential of callus derivedfrom male inflorescence decreased while the calli were subcultured over 3 times with the internalof more than 3 weeks. The frequency of regenerating shoot ranged from 31.8%to 35.0%inmedium containing 1.0 mg/L 6-BA under the dim light. The addition of 3%maltose intoregeneration medium not only could alleviate browning,but also contributed to regenerate shoot.The rooting rate of shoots was 62.5%in 1/2 MS medium containing 0.3 mg/L IBA.
