水稻程序性细胞死亡抑制基因Dad-1在薏苡中的定位

Chromosomal Localization of Programmed Cell Death Suppressor OsDad-1 Gene in Coix lacryma-jobi L.

  • 摘要: 基因Dad-1是普遍存在于动物和植物中一个高度保守的程序性细胞死亡抑制基因。以水稻Dad-1基因为探针,采用Southern杂交在薏苡中检出了Dad-1基因的同源序列,并利用荧光原位杂交的方法对其进行了染色体物理定位。在薏苡第9染色体短臂上检测到Dad-1基因的杂交信号,信号与着丝粒的百分距离为67.44±1.45。

     

    Abstract: The gene Dad-1 is a well-conserved programmed cell death (PCD) suppressor gene in animals and plants.In this study,the single copy rice gene Dad-1 was used as probe to investigate its homology and physical localization in Coix lacryma-jobi genome.The results of Southern blotting suggested that there were sequences homologous to rice Dad-1 gene in C.lacryma-jobi genome.Further,the sequences were mapped on the short arm of chromosome 9 in C.lacryma-jobi by fluorescence in situ hybridization (FISH).The percent distance from the signal site to centromere was 67.44±1.45 .
